The University of Michigan Transgenic Animal Model Core will offer a CRISPR/Cas9 and Mouse ES Cell Training Class beginning Monday, November 4 through Friday, November 15.

In this two-week hands-on session, trainees will design, build, and validate CRISPR/Cas9 reagents in mouse embryonic stem (ES) cells.

Led by course instructors Thom Saunders, PhD, and Elizabeth Hughes, MS, trainees will learn how to work with mouse embryonic stem cells in culture, to clone CRISPR/Cas9 reagents in plasmids, and to purify and analyze DNA from cultured cells.

Trainees will learn fundamental genetic principles for genetic engineering including: 

  1. Planning CRISPR/Cas9 genome editing workflows,
  2. Fundamental principles of gene targeting design, and
  3. How to apply Cre recombinase technology to conditional genes (“floxed genes”).

In addition, trainees will be introduced to the power of genetically engineered mice to model human disease and learn how to access resources from the International Knockout Mouse Consortium project to knockout every protein-coding gene in the mouse.

After attending, trainees will be able to design experiments to create genetic modifications and produce them in mouse ES cells and in mice with CRISPR/Cas9 reagents.
